The ability of the cellulolytic complex obtained from Trichoderma reesei QM 9414 grown on Solka floc, to hydrolyze the lignocellulosic biomass of Onopordum nervosum Boiss native or pretreated by alkali is studied. Some properties of that cellulolytic complex were previously tested in order to determine the best conditions to carry out the enzymatic hydroysis experiments. Optimum enzymatic activity has been found when hydrolysis was performed at 50oC and pH from 4.3 to 4.8, using 0.05 M citrate buffer or 0.1 M acetate buffer. Studies on thermal stability at 50oC over a 96 hour incubation period have shown an filter paper and B-glucosidase activity respectively. Enzymatic hydrolysis experiments were carried out by using Onopordum nervosum, untreated or pretreated with alkali, at a 5. SUMMARY. substrate concentration and with a filter paper activity about 20 FPU per gram of substrate. The hydrolysis yield after a 48 hour hydrolysis period showed an increase of more than three times, based on dry weight of substrate, by effect of the alkaline pretreatment. Final cellulose to glucose conversion, taking into account the glucose solubilization during the pretreatment, was 160. INTRODUCTION. higher than that obtained for the untreated substrate.
